becton dickinson certificate of analysis: Köhler's Invention Klaus Eichmann, 2005-07-19 Georges Köhler was one of the most prominent German scientists of recent history. In 1984, at an age of 38, he received the Nobel Prize in Physiology or Medicine, together with N.K. Jerne and C. Milstein, for inventing the technique for generating monoclonal antibodies. This method and its subsequent applications had an enormous impact on basic research, medicine and the biotech industry. In the same year, Köhler became one of the directors of the Max-Planck-Institute of Immunobiology in Freiburg; his unfortunate premature death in 1995 set an end to his extraordinary career. Prof. Klaus Eichmann, who had invited Köhler to become his codirector, is one of the people who were closest to him. This scientific biography commemorates the 10th anniversary of Köhler's untimely death. Köhler's scientific achievements are explained in a way to make them understandable for the general public and discussed in the historical context of immunological research. |

Mulay, Martin Herrmann, Rostyslav Bilyy, Alexander Gabibov, Hans-Joachim Anders, 2019-07-19 Nano- and microparticles including crystals, synthetic biomaterials, misfolded proteins or environmental particulates are involved in a wide range of biological processes and diseases. They may present as intrinsic or environmental toxins but may also be applied intentionally, e.g. as immune adjuvants, drug carriers or ion exchangers. The discovery that a wide range of nano- and microparticles share the capacity to induce IL-1β secretion via activation of the NLRP3 inflammasome in dendritic cells and macrophages has led to the hypothesis that nano- and microparticles may contribute in a uniform mechanistic manner to different disease entities. Other molecular mechanisms triggered by a range nano- and microparticles have also recently been identified including (i) the induction of regulated necrosis; (ii) neutrophil extracellular trap (NET) formation and (iii) foreign body granuloma formation as a mechanism of persistent tissue inflammation and scarring. Research on the biology of nano- and microparticle handling is currently under intense investigation. The cell type-specific responses of nano- and microparticle exposure deserves careful attention as well as the related secondary responses to these particles that lead to tissue remodeling. The immune system is at the center of these processes in terms of particle clearance, particle-induced cell death and inflammation, thereby limiting particle-related inflammation and orchestrating wound healing responses. In this Research Topic, we welcomed the submission of Original Research, Review and Mini-Review articles that addressed the significance of the immune system in particle-induced cell death, inflammation and immune responses. These findings will help facilitate new approaches to the prevention and management of particle-related diseases. |

Meade, 2015-07-22 The world’s population is predicted to hit 9 Billion by 2050, and with it food demand is predicted to increase substantially. The World Bank estimates that cereal and meat production needs to increase by 50% and 85% respectively between 2000 and 2030 to meet demand, putting serious pressure on the global agricultural industry. Critical to meeting this demand for food are mechanisms to reduce the incidence of animal disease. With in excess of 1.3 billion cattle globally, the total cost of infectious diseases is difficult to estimate. However in North America alone, the cost is predicted to be $18 billion annually. Non-infectious diseases also account for another major impediment to the production capacity and welfare of animals as well as the economic sustainability of farming. However animal diseases have implications that spread far beyond the farm gate. Infectious agents can also contaminate the food chain, and potentially affect human health. Controlling diseases, through better preventative and treatment methods requires a detailed understanding of the immune response in livestock species. Multiple studies have identified associations between variation in immune genes and disease susceptibility, which potentially opens up new avenues to select animals with superior disease resistance. Detailed understanding of immunity in cattle is leading to the design of more effective vaccines. Furthermore, appreciation of the significant differences between rodent and human immune responses has also led to bovine models being developed for some human diseases. The publication of the bovine genome and the advent of next-generation sequencing technologies have facilitated a massive expansion in our knowledge of the immune response in cattle. As a result there has been an explosion of exciting research findings including in metagenomics and epigenetics. Recently, there has been a welcome move to integrate our emerging understanding of the immune response with detailed studies of other important physiological processes including nutrition and reproduction. The interactions between the reproductive system, nutrition and the immune system are of particular interest, since each places significant demands on the animal at various stages through the production cycle. The interplay between these morphologically diffuse systems involves widely distributed chemical signals in response to environmental input, and each system must interact for the normal functioning of the other. A comprehensive “systems” approach is improving our understanding of normal physiological interactions between these systems and furthermore, how dysregulation can lead to disease. The successful translation of bovine immunological research into improved treatments for animal disease requires tight interaction between diverse scientific and clinical disciplines including immunology, microbiology, endocrinology, physiology, nutrition, reproduction and clinical veterinary medicine. With so much recent progress in the field, we believe that it is valuable and well-timed to review the broad variety of the relevant studies that attempt to increase our understanding through comprehensive collaboration between these disciplines. Abedon, Pilar García, Peter Mullany, Rustam Aminov, 2017-09-05 Historically, the first observation of a transmissible lytic agent that is specifically active against a bacterium (Bacillus anthracis) was by a Russian microbiologist Nikolay Gamaleya in 1898. At that time, however, it was too early to make a connection to another discovery made by Dmitri Ivanovsky in 1892 and Martinus Beijerinck in 1898 on a non-bacterial pathogen infecting tobacco plants. Thus the viral world was discovered in two of the three domains of life, and our current understanding is that viruses represent the most abundant biological entities on the planet. The potential of bacteriophages for infection treatment have been recognized after the discoveries by Frederick Twort and Felix d’Hérelle in 1915 and 1917. Subsequent phage therapy developments, however, have been overshadowed by the remarkable success of antibiotics in infection control and treatment, and phage therapy research and development persisted mostly in the former Soviet Union countries, Russia and Georgia, as well as in France and Poland. The dramatic rise of antibiotic resistance and especially of multi-drug resistance among human and animal bacterial pathogens, however, challenged the position of antibiotics as a single most important pillar for infection control and treatment. Thus there is a renewed interest in phage therapy as a possible additive/alternative therapy, especially for the infections that resist routine antibiotic treatment. The basis for the revival of phage therapy is affected by a number of issues that need to be resolved before it can enter the arena, which is traditionally reserved for antibiotics. Probably the most important is the regulatory issue: How should phage therapy be regulated? Similarly to drugs? Then the co-evolving nature of phage-bacterial host relationship will be a major hurdle for the production of consistent phage formulae. Or should we resort to the phage products such as lysins and the corresponding engineered versions in order to have accurate and consistent delivery doses? We still have very limited knowledge about the pharmacodynamics of phage therapy. More data, obtained in animal models, are necessary to evaluate the phage therapy efficiency compared, for example, to antibiotics. Another aspect is the safety of phage therapy. How do phages interact with the immune system and to what costs, or benefits? What are the risks, in the course of phage therapy, of transduction of undesirable properties such as virulence or antibiotic resistance genes? How frequent is the development of bacterial host resistance during phage therapy? Understanding these and many other aspects of phage therapy, basic and applied, is the main subject of this Topic. |
